The relationship between efferents of the hypothalamic suprachiasmatic nucleus SCN and neurons of the thalamic paraventricular nucleus PVT projecting to the amygdala was investigated in the rat using tract tracing in light and electron microscopy. Biotinylated dextran amine was used to label anterogradely SCN efferents.

The size, shape and cellular morphology of the suprachiasmatic SCN and paraventricular nuclei PVN in the human hypothalamus were examined in relation to sex and age. In both nuclear regions the following parameters were determined: length of the rostrocaudal axis, maximum cross sectional area, volume, numerical cell density, total number of cells and the diameter of cell nuclei. In contrast to the absolute measurements, sexual differences were found in the internal structural organisation of these hypothalamic nuclei using multivariate regression analysis.

Physiological set points refer to the baseline level at which functions such as heart rate, and at which chemical compositions such as plasma sodium concentration are normally maintained. These set points are represented in the brain by specific discharge rates in neurons dedicated to the monitoring and control of specific physiological processes. Thus, separate groups of neurons are dedicated to the control of heart rate, temperature, etc. The hypothalamus has the greatest concentration of nuclei at which set points are encoded, monitored and controlled, and so can be considered as the key brain region for the control of homeostasis.

The suprachiasmatic nucleus or nuclei SCN is a tiny region of the brain in the hypothalamus , situated directly above the optic chiasm. It is responsible for controlling circadian rhythms. The neuronal and hormonal activities it generates regulate many different body functions in a hour cycle. The mouse SCN contains approximately 20, neurons. The SCN interacts with many other regions of the brain. It contains several cell types and several different peptides including vasopressin and vasoactive intestinal peptide and neurotransmitters. The paraventricular nucleus of the hypothalamus PVH , located in the ventral diencephalon adjacent to the third ventricle, is a highly conserved brain region present in species from zebrafish to humans. The PVH is composed of three main types of neurons, magnocellular, parvocellular, and long-projecting neurons, which play imperative roles in the regulation of energy balance and various endocrinological activities. In this review, we focus mainly on recent findings about the early development of the hypothalamus and the PVH, the functions of the PVH in the modulation of energy homeostasis and in the hypothalamus-pituitary system, and human diseases associated with the PVH, such as obesity, short stature, hypertension, and diabetes insipidus. Thus, the investigations of the PVH will benefit not only understanding of the development of the central nervous system but also the etiology of and therapy for human diseases.
